Antigen: Dog IgE.

Monoclonal Designations: E6-2A1, E6-59A1, and E6-71A1.

Ig Isotype/subclass plus light chain: E6-2A1 = IgG2A kappa , E6-59A1 = IgG1 kappa E6-71A1 = IgG1 kappa.

Specific reactivity: Specific for Dog IgE; these MAbs do not react with dog IgM; IgA, or IgG1 or IgG2. Comments: There may be two dog IgE subclasses, if so these MAbs are likely specific for both IgE subclasses. These MAbs are closely related, but each is different.

Properties:

Western blot: with affinity purified dog IgE in reducing gels each MAb gives one major band at 65-62 kD.

ELISA: Each of the 3 E6- MAbs will perform as a specific capture MAb, and each (when biotinylated) will also perform as an ELISA probe for dog IgE levels in serum. The more effective combinations are E6-2 as capture or probe with E6-71 in apposition. Affinity purification: Both E6-2A1 or E6-71A1 (when cross-linked to a column matrix) are effective in specifically isolating dog IgE from serum. Detection of specific allergens: Each of the MAbs will detect housemite specific dog IgE in an ELISA format.

Skin sensitivity: Each MAb will rapidly induce large immediate hypersensitivity wheals if injected into dogs i.d. in ug. or ng. quantities.

Preparation and supply: SpA purified from tissue culture supernatant and supplied in PBS pH 7.2 with 0.05% sodium azide as preservative. Biotin conjugated MAb also usually available.
